cDNA Library Construction. Double-stranded cDNA was synthesized with SuperScriptII RT and DNA polymerase I and ligated to adapters containing a SalI restriction site (GIBCO/BRL). To remove residual adapters and enrich for full-length fragments, cDNA was first size-fractionated by column chromatography and then ligated to predigested pSportI vector (GIBCO/BRL). Transformation of DH5α with the ligated DNA yielded a plasmid library with approximately 8.8 x 10 4 colony-forming units.
